After once or twice freeze-thaw cycle, JetGiga DH5α shows high transformation efficiency.

Protocol

6-minute Protocol with Higher Transformation Efficiency (>1×109 CFU/μg)

6 minutes Procedure of JetGiga

(1) Thaw JetGiga DH5α on ice.
(2) Add plasmid DNA to the cells and mix gently by tapping.
(3) Incubate the cells on ice for 5 minutes.
(4) Incubate the cells in a 42℃ water bath for 30 seconds.
(5) Spread it on selection plates.

10-minute Heat-Shock-Free Protocol with High Transformation Efficiency (>1×108 CFU/μg)

10 minutes Procedure of JetGiga

This protocol is suitable for automation due to the absence of a heat-shock step.

(1) Thaw JetGiga DH5α on ice.
(2) Add plasmid DNA to the cells and mix gently by tapping.
(3) Incubate the cells on ice for 5 minutes.
(4) Add Recovery Medium to the tube and vortex it for 1 second.
(5) Incubate the cells at room temperature for 5 minutes.
(6) Spread it on selection plates.
